Description
Curcumin has generated tens of thousands of publications, which makes it one of the most studied natural compounds in existence. It is also the subject of a well-known methodological critique: it behaves as a pan-assay interference compound, producing apparent activity across an implausibly wide range of screening assays. Both the volume of research and that critique are essential context for reading its literature.
Mechanisms of Action
Curcumin is a diarylheptanoid polyphenol from Curcuma longa. Research describes inhibition of NF-kB signalling with downstream reduction in inflammatory cytokine transcription, modulation of COX-2 and lipoxygenase pathways, and direct radical scavenging activity. Studies also document extremely poor aqueous solubility, rapid glucuronidation and low systemic bioavailability, which is why most research formulations employ delivery enhancement. The compound's tendency to produce non-specific signals in screening assays is well documented in the analytical literature.

An enormous but methodologically contested literature. The volume of publications considerably exceeds the volume of well-controlled research, and assay interference is a documented confounder.
