Nicotinamide riboside has the strongest human evidence base of the NAD+ precursors — not necessarily the strongest effects, but the most trials, the most pharmacokinetic data and the clearest regulatory position. It enters the salvage pathway a step further upstream than NMN, which gives it a different and better-characterised transport profile. That combination of reliable pharmacokinetics and more variable functional findings makes it the most useful comparator in precursor research.
Mechanisms of Action
NR is a pyridine nucleoside converted to NMN by nicotinamide riboside kinases, and subsequently to NAD+ by NMNAT enzymes. Research describes cellular uptake via equilibrative nucleoside transporters, a well-characterised route that distinguishes it from the contested transport picture for NMN. Human pharmacokinetic studies consistently document elevation of whole blood NAD+ following administration. Studies also report substantial first-pass conversion to nicotinamide, which is a recognised consideration in interpreting tissue-level effects. The relative efficiency of the three principal precursors remains an open comparative question.
