Description
SLU-PP-332 is a synthetic agonist of the estrogen-related receptor family, a group of orphan nuclear receptors central to oxidative metabolism. Research interest stems from the observation that these receptors sit upstream of the transcriptional programmes governing mitochondrial biogenesis and fuel selection. The compound is recent, entirely preclinical, and its literature is dominated by the group that developed it.
Mechanisms of Action
SLU-PP-332 is reported to act as a pan-agonist at estrogen-related receptors alpha, beta and gamma. These receptors function as transcriptional regulators of genes governing oxidative phosphorylation, fatty acid oxidation and mitochondrial biogenesis, working in concert with PGC-1alpha. Research in skeletal muscle models describes upregulation of oxidative gene programmes and increased fatty acid utilisation, with reported changes in exercise capacity in rodent studies. The compound is a small molecule rather than a peptide and its pharmacokinetic profile is not well characterised.
Research Status & Findings
A very small and recent literature confined to rodent and cell models, produced largely by a single research group. Independent replication is limited and no human data exist.
