PNC-27 – 5mg (3ml Pen)

PNC-27 is an experimental research construct combining a p53-derived sequence with a membrane-penetrating domain. Its research premise is selectivity: that the construct interacts with a protein complex expressed on the surface of certain transformed cells but not on normal cells. The literature is small, preclinical, and largely produced by the original research group, which is an important limitation on how the findings are read.

PNC-27 combines residues from the p53 HDM-2 binding domain with a penetratin-derived membrane-residency sequence. Research describes binding to HDM-2 expressed in the plasma membrane of cancer cell lines, followed by pore formation and membrane disruption leading to necrotic cell death. Studies report that the proposed selectivity derives from differential membrane HDM-2 expression between transformed and untransformed cells. The mechanism described is membrane-lytic rather than dependent on intracellular p53 pathway restoration.

A small preclinical literature confined to cell line and limited animal work, produced predominantly by a single research group. Independent replication is sparse and no clinical data exist.
